Description
The CRISPRiTest™ assay measures the transcriptional repressor function of any dCas9-repressor fusion, such as Cellecta's dCas9-KRAB variant, or dCas9 itself in any mammalian cell system. Cellecta's Functional Cas9/dCas9 Fluorescent Reporter Assays work differently than Cellecta's Cas9 Essential Gene Assays (i.e., the CRISPRtest kits). While readout for both types of Cas9 assays relies on the the disappearance of a fluorescent marker to assess Cas9 activity, the Essential-Gene Cas9 relies on cell death induced by disruption of a gene required for viability, whereas, with the CRISPR-test Fluorescent Reporter Assays, the sgRNA directly interferes with expression of one of the reporter genes. As a result, the Cellecta Functional Cas9/dCas9 Fluorescent Reporter Assays can be used with any mammalian cell system. Each kit provides enough reagents to measure 5-10 cell lines
